Do Differentiated Osteoblasts and Allogeneic Mesenchymal Stromal Cells Augment Bone Formation?

چکیده
Introduction: Fixation of massive bone tumour endoprosthetic replacements is not as successful as conventional joint replacement surgery and aseptic loosening is the major cause of implant failure. Increased bone growth and contact to the implan shaft by bony bridging has been shown vital in reducing implant failure. Our previous work has shown that spraying autogenic bone marrow derived mesenchymal stromal cells (MSCs) within fibrin glue onto the grooved hydroxyapatite (HA) collars of segmental bone tumour implants significantly increased bone formation in an ovine model1. In this study we investigated the hypothesis that allogeneic MSCs and MSC’s differentiated in an osteogenic supplemented media (ogMSCs) will improve bone growth equal to undifferentiated autogenic MSCs in an in vivo massive tumor implant ovine model. This study also hypothesized that bone growth is dependent on cell concentration.

The effect of bone marrow-derived mesenchymal stem cells to induce PD-L1 molecule on splenic lymphocytes
Background: Mesenchymal stem cells are non-hematopoietic stromal cells that are used in the treatment of many chronic and autoimmune diseases by modulating the immune system. Due to the limitations of using autologous mesenchymal stem cells, the use of allogeneic stem cells is a promising therapeutic approach in the treatment of immunological disorders. This study aimed to investigate the abili...
متن کاملAn animal model study for bone repair with encapsulated differentiated osteoblasts from adipose-derived stem cells in alginate
Objective(s): Adipose derived stem cells (ADSCs) can be engineered to express bone specific markers. The aim of this study is to evaluate repairing tibia in animal model with differentiated osteoblasts from autologous ADSCs in alginate scaffold. Materials and Methods: In this study, 6 canine’s ADSCs were encapsulated in alginate and differentiated into osteoblasts. Alkaline phosphatase assay (A...
متن کاملReconstitution of bone-like matrix in osteogenically differentiated mesenchymal stem cell–collagen constructs: A three-dimensional in vitro model to study hematopoietic stem cell niche
Mesenchymal stem/stromal cells (MSCs) and osteoblasts are important niche cells for hematopoietic stem cells (HSCs) in bone marrow osteoblastic niche. Here, we aim to partially reconstitute the bone marrow HSC niche in vitro using collagen microencapsulation for investigation of the interactions between HSCs and MSCs. Mouse MSCs (mMSCs) microencapsulated in collagen were osteogenically differen...
متن کاملInduction of Mineralized Nodule Formation in Rat Bone Marrow Stromal Cell Cultures by Silk Fibroin
Background: Silk fibroin is a suitable protein for osteogenesis by inducing markers of bone formation in the cultures of osteoblasts, so we examined the ability of this protein to induce mineralized nodules in the rat bone marrow stromal cell cultures. Methods: Bone marrow stromal cells obtained from 4 to 6 weeks old Spruge-Dawely male rats were grown in primary culture for seven days and then ...
متن کاملCapillary Network Formation by Endothelial Cells Differentiated from Human Bone Marrow Mesenchymal Stem Cells
Human bone marrow derived mesenchymal stem cells (HBMSCs) have the potential to differentiate into cells such as adipocyte, osteocyte, hepatocyte and endothelial cells. In this study, the differentiation of hBMSCs into endothelial like-cells was induced in presence of vascular endothelial growth factor (VEGF) and insulin-like growth factor (IGF-1). The differentiated endothelial cells were exam...
